將培養後的細菌進行核酸粗萃取後，利用聚合酶連鎖反應來進行鑑定結核桿菌群。在巢式聚合酶連鎖反應過程中，目標核酸片段會標記上特定標定物並擴增，再將其擴增產物滴到免疫層析檢測試紙卡匣內，待五分鐘後，即可讀取結果。
